Do Estate managers have specials rates on setup land and maintenance fees...if yes what are they?

I want to know that because I am interested to open an estate, but I want to know if the estate owners have some more bennefits over regular peoples, to make the business possible. ty.

Usually no.  Some very large customers (allegedly in the order of 50 sims or more @ US$195 = US$9,750/month) have got special rates from LL but in general the business is possible because you have to be premium to rent land from LL and many residents are not - so they have to rent from estate owners if they want land at all.

So, if you're going to be investing US$10k per month or more have a word with LL.

You do not need to be a premium member to buy a private estate, you will get Concierge level support which is a step up from premium level. You can also get concierge support if you own more than half a sim of mainland, but you would also need premium membership for that.

If you pay more than 5K USD per month in tier fees, you would then get "Extended support" which entitles you to sim transfers within 48 hours and tickets resolved within 24 hours.
