Land holdings are always paid monthly, so if you purchase 1/2 sim on March 3, yes you will be billed 125 USD, regardless of your premium account billing rate and date. It may be worth a call to the billing support (or contact Live Chat) to check this out with them.
In reply to your second question: LL always charges your USD balance, not your credit card directly. It's up to you to increase your credit before your due date. Having a CC registered does not entitle LL to charge your bank account, so you have to be careful to have enough on your USD balance for each due date.
