So a bit of land I want to buy is part of an 8192m chunk, I have premium plus so 1/4 of that is effectively free, or at least cost factored into my membership.

And then I realistically only intend to use about half of it at most? So I’d probably sell half

But let’s say it sells within a week of buying the original 8192m bit of land. And I’m down to 4096 or effectively 2048 worth of tier. How would it be calculated and billed? As if I had 6144m straight after purchase even if 4096 is already sold? Or would it be as if it were 2048m?
I’m not super concerned about the cost difference, just kind of wondering how the billing timing works.

In general unless things have changed you pay tier on the LARGEST amount of land you own in that tier  pay period. You can find your official tier date in your back end (it is not necessarily the date you bought the current land but was anyway based on the FIRST TIME you bought land).

You can find a land calculater in your web account backend also and there is a LAND wiki. All good resources.  And yes, it is very confusing and tricky and  takes many folks a while to "get" how it works so explaining id difficult at best. Hence no one jumping in to answer this.  

Even if you own the land for only a minute it counts.  So calculate, buy and sell carefully.   

You can ask support but when I did that long ago I was given an incorrect answer and ended up paying more than I wanted to --- so there is that LOL.   

Sure someone else can explain better. I have been renting now for a long time.

This has been my experience recently (highest amount during the period). Your land use fee level doesn't automatically drop if you sell land to move to a lower level, which makes sense, LL doesn't know whether you plan to acquire more. If you want to drop down a level, manually check the box under Land Use Fees.

the others explained the things about holding right, also be indeed sure to check your tier level after cutting down.

BUT the time.. you will even with one second owning a large parcel IN your monthly billing cycle... pay the full amount for that tier. Tier is after all, paid afterwards and not as rent upfront. The next cycle will be the first one to drop.

The billing cycle for land started the moment you bought your first mainland, that date will not change, and is mostly not the same as your premium paying date.
