You can keep yourself on busy mode all the time, but that also means you won't get group notifcations or recieve any items you purchase while in busy mode, so it's a double-edged sword. There is no way to prevent people from asking to be your friend; that part's on their side. You can only individually decline any friendship offers. You could also put something in your profile about it, so anyone who bothers to read will see that you don't add friends.

You don't want any friends??  Well, the others have some great advice, but mine is more general.

Second Life is a social medium (as well as being many other things).  You can't take that social aspect out of SL, no matter how much of a grumpy old misanthrope you are.  But you can have your very own personal virtual world, in which you are the only inhabitant and will never have to worry about interacting with others or their bothersome attempts to be friendly.  Just download and install the open source virtual world software OpenSim on your computer.

just on a funny note: if you don't mind you can also change your display name to :donotwantfriends

But even this does not prevent you from having some very optimistic persons still trying to add you and hoping to win you and your grumpy personality as a friend.

As the others already stated, best is to decline the offers, or even go with Lindal's advice and create your very own local region on your computer with OpenSIM, there certainly no one will ever disturb you. ; )
