That all depends on the store.  Just like RL, each store has a different way of doing it.  You can usually find a sign in the in-world store that tells you how to use it, though.  If not, contact the merchant and ask.

Also, try rezzing it (drag the giftcard box from your inventory on to the ground in a sandbox, then right-click it and open. copy contents back to your inventory). Sometimes if you 'wear' the giftcard briefly you will activate it. It should let you know in a pop-up window once it's activated. Then when you purchase an item from a vendor poster in the store it will tell you that you have store credit to use up...
